  WB analysis of fetal human brain using 66282-1-Ig

  Untreated and PNGase F-treated lysates of fetal human brain tissue and human spleen tissue were subjected to SDS PAGE followed by western blot with 66282-1-Ig (CD200 antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. PNGase F was obtained from Atagenix (cat.NO. ata808).

  1X10^6 U-937 cells were stained with 0.2 ug Anti-Human CD200 (66282-1-Ig, Clone: 5F3D6) and CoraLite®488-Conjugated AffiniPure Goat Anti-Mouse IgG(H+L) at dilution of 1:1000 (green), or with 0.2 ug mouse IgG1 isotype control antibody and CoraLite®488-Conjugated AffiniPure Goat Anti-Mouse IgG(H+L) at dilution of 1:1000 (black). Cells were fixed with 90% MeOH.

Background Information

CD200, also known as OX2, is a type I transmembrane glycoprotein that belongs to the immunoglobulin superfamily (PMID: 3032785). It contains two extracellular immunoglobulin domains, a transmembrane and a cytoplasmic domain. CD200 is expressed on a variety of cell types, including thymocytes, B lymphocytes, a subset of T lymphocytes, follicular dendritic cells, neurons, and endothelial cells (PMID: 3032785; 10981966). CD200 binds to its receptor, CD200R, which is primarily expressed by myeloid and T cell lineage (PMID: 15187158). The CD200-CD200R interaction plays a role in immunosuppression and suppression of anti-tumor immune responses (PMID: 22020332).
