  WB analysis using 87669-1-RR (same clone as 87669-1-PBS)

  HEK-293T cells and mouse Cxcl12 transfected HEK-293T cells lysates were subjected to SDS PAGE followed by western blot with 87669-1-RR (CXCL12/SDF-1 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 87669-1-PBS in a different storage buffer formulation.

  IHC staining of mouse spleen using 87669-1-RR (same clone as 87669-1-PBS)

  Immunohistochemical analysis of paraffin-embedded mouse spleen tissue slide using 87669-1-RR (CXCL12/SDF-1 antibody) at dilution of 1:1000 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 87669-1-PBS in a different storage buffer formulation.

  Biolayer interferometry (BLl) kinetic assays of 87669-1-RR against Mouse CXCL12/SDF-1 were performed. The affinity constant is 4.03 nM.

Background Information

In adulthood, CXCL12 plays an important role in angiogenesis by recruiting endothelial progenitor cells (EPCs) from the bone marrow through a CXCR4 dependent mechanism (PMID: 17878755). It is this function of CXCL12 that makes it a very important factor in carcinogenesis and the neovascularisation linked to tumour progression (PMID: 16943240). CXCL12 also has a role in tumor metastasis where cancer cells that express the receptor CXCR4 are attracted to metastasis target tissues that release the ligand, CXCL12 (PMID: 11242036 ). In breast cancer, however, increased expression of CXCL12 determines a reduced risk of distant metastasis (PMID: 19646861; 17724466 ).
