Background Information
Profilaggrin is a major protein component of the keratohyalin granules of mammalian epidermis. It is initially expressed as a large polyprotein precursor, which is subsequently proteolytically processed into individual functional filaggrin molecules. The free filaggrin binds to keratin intermediate filaments, causing their aggregation into macrofibrils in which the intermediate filaments are aligned in tightly packed parallel arrays. Mutations in Filaggrin are associated with Ichthyosis vulgaris and Dermatitis atopic 2.(PMID: 19386895, 16550169)
