  FC experiment of rat splenocytes using 30293-1-AP

  1X10^6 PMA and ionomycin treated rat splenocytes cells were intracellularly stained with 0.5 ug Anti-Rat IFN Gamma (30293-1-AP) and CoraLite®488-Conjugated AffiniPure Goat Anti-Rabbit IgG(H+L) at dilution 1:1000, and 0.5 ug APC Anti-Rat CD4 (OX-35) (APC-65179, Clone: OX-35), and 0.5 ug Rabbit IgG control Rabbit PolyAb (30000-0-AP, Clone: ). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer (PF00011-C).

Tested Applications

• Positive IHC detected in: rat spleen tissue; Note: suggested antigen retrieval with TE buffer pH 9.0; (*) Alternatively, antigen retrieval may be performed with citrate buffer pH 6.0
• Positive FC detected in: PMA and ionomycin treated rat splenocytes cells

Recommended dilution

• Immunohistochemistry (IHC): IHC : 1:50-1:500
• Flow Cytometry (FC): FC : 0.50 ug per 10^6 cells in a 100 µl suspension

Background Information

IFN gamma (IFNG) is a soluble cytokine that is the only member of the type II class of IFN. It is secreted by Th1 cells, cytotoxic T cells and NK cells. The cytokine is associated with antiviral, immunoregulatory and anti-tumor properties and is a potent activator of macrophages. It plays crucial roles in pathogen clearance. Aberrant IFNG expression is associated with a number of autoinflammatory and autoimmune diseases. It has been identified in many studies as a biomarker for pleural tuberculosis (TB).
