Background Information

ITGB1BP2 (Integrin Subunit Beta 1 Binding Protein 2), also known as Melusin, is a protein-coding gene located on the X chromosome (Xq13.1). It encodes a 347-amino acid protein that is highly conserved, sharing 92% sequence identity with its mouse homolog. The protein is characterized by two cysteine- and histidine-rich (CHORD) domains, PXXP motifs, YXXI/P motifs, putative SH2 and SH3 domain binding motifs, and a C-terminal acidic region that resembles the calcium-binding domains of calreticulin and calsequestrin. Functionally, ITGB1BP2 interacts specifically with the cytoplasmic domain of the beta-1 integrin subunit and is thought to act as a chaperone protein. Its expression is highly restricted, being detected predominantly in skeletal and cardiac muscles, and it is believed to play a crucial role in the maturation and organization of muscle cells. Notably, studies in mouse models have demonstrated that the absence of this gene leads to a failure in cardiac hypertrophy in response to mechanical stress, highlighting its essential role in the heart's adaptive response to biomechanical load.
