  LNCaP cells were subjected to SDS PAGE followed by western blot with 84059-5-RR (KLK3/PSA antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 84059-5-PBS in a different storage buffer formulation.

  Immunohistochemical analysis of paraffin-embedded human prostate cancer tissue slide using 84059-5-RR (KLK3/PSA antibody) at dilution of 1:500 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 84059-5-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (4% PFA) fixed LNCaP cells using KLK3/PSA antibody (84059-5-RR, Clone: 241263F8 ) at dilution of 1:250 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2). This data was developed using the same antibody clone with 84059-5-PBS in a different storage buffer formulation.

  FC experiment of HeLa using 84059-5-RR (same clone as 84059-5-PBS)

  1x10^6 HeLa cells were intracellularly stained with 0.25 ug KLK3/PSA Recombinant antibody (84059-5-RR, Clone:241263F8) and PE-conjugated Goat Anti-Rabbit IgG(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with True-Nuclear Transcription Factor Buffer Set. This data was developed using the same antibody clone with 84059-5-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 84059-5-RR

  Biolayer interferometry (BLl) kinetic assays of 84059-5-RR against Human KLK3/PSA were performed. The affinity constant is 149 nM.

Background Information

KLK3 also known as Prostate-Specific Antigen (PSA), is a 33 kDa glycoprotein primarily synthesized by the epithelial cells of the prostate gland. Its main physiological function is to cleave seminogelins I and II in semen, leading to the liquefaction of the seminal coagulum and the release of motile sperm. It is under tight regulation by androgens via the androgen receptor (AR). Serum PSA levels are elevated in prostate cancer due to disruption of the glandular architecture, allowing more PSA to enter the bloodstream. It is used alongside digital rectal exam (DRE) for early detection. Although termed "prostate-specific," PSA is not exclusively produced by the prostate. Very low levels can be produced in periurethral glands, breast tissue, and some other tissues. Its expression is highly abundant in prostatic tissue and seminal fluid.
