Background Information
Niemann-Pick Type C (NPC) disease is a lysosomal storage disorder characterized by the accumulation of unesterified cholesterol and other lipids in the endolysosomal system. NPC disease results from a defect in either of two distinct cholesterol-binding proteins: a transmembrane protein, NPC1, and a small soluble protein, NPC2. NPC1 or NPC2 deficiency models showed that the functions of these two proteins within lysosomes are closely linked. NPC1 is a typical transmembrane protein and contains a number of modification sites for glycosylation. Defects in NPC1 are the cause of Niemann-Pick disease type C1, which exhibits a highly variable clinical phenotype. Moreover, NPC1 may play a role in vesicular trafficking in glia, a process that may be crucial for maintaining the structural and functional integrity of nerve terminals.(PMID: 20525256; PMID: 31608980; PMID: 15465421)

Reviews
What customers say
All three reviews are positive, with ratings of 5, 5, and 4 out of 5. Users report reliable and specific detection of NPC1 across multiple applications. One user found excellent specific staining in immunofluorescence on MEF cells at 1:200 dilution. Two users confirmed clear Western Blot results at 1:1000 dilution—one in HAP1 whole cell lysate and another in primary mouse neurons—both showing the band at the correct molecular weight. Overall, reviewers highlight the antibody's specificity, consistent performance, and suitability for both IF and WB.
The reviews below have been submitted by verified Proteintech customers who received an incentive for providing their feedback.
Parijat (Verified Customer) (04-08-2024) | Good antibody for specific staining of NPC1 in IF.
|
Ganesh (Verified Customer) (02-07-2024) | 25 µg of HAP1 (human) whole cell lysate was resolved on a 4-12% Bis-Tris Gel and proteins were transferred onto a nitrocellulose membrane. NPC1 antibody was diluted at 1:1000 in Blocking Buffer (5% Milk in 1x TBS-T (0.1% Tween-20)) and the membrane was incubated overnight at 4 deg C on a rotator.
