We tested this antibody as an alternative to αSMA for the identification of pericytes by immunohistochemistry in murine tumor tissues with strong angiogenesis. To optimize staining, we performed several conditions. Initial testing was done in colon tissue (32 min antigen retrieval, 1:100 dilution, 40 min incubation). We then tested placenta and testis with 40 min antigen retrieval and 1:100 dilution. Additional experiments were performed using 40 min antigen retrieval with a higher concentration (1:50) in kidney, placenta, and colon. Finally, we tested 40 min antigen retrieval with 1:50 dilution and overnight incubation in kidney, placenta, and colon. Despite testing multiple antigen retrieval conditions, antibody concentrations, and incubation times, we were unable to detect specific staining in any of the tested tissues. Under our experimental conditions, the antibody did not produce detectable labeling by immunohistochemistry, at least in our hands.
