  WB analysis of HEK-293 using 80431-2-RR (same clone as 80431-2-PBS)

  Non-treated HEK-293 cells, phosphatase inhibitor treated HEK-293 cells and λ phosphatase treated HEK-293 cells were subjected to SDS PAGE followed by western blot with 80431-2-RR (Phospho-BAD (Ser112) antibody) at dilution of 1:1000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with GAPDH (60004-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 80431-2-PBS in a different storage buffer formulation.

Background Information

BAD, a distant member of the BCL-2 family, exerts its death promoting e€ect by hetero-dimerizing with BCL-XL and BCL-2 death antagonists in the mitochondria. The function of the pro-apoptotic molecule BAD is regulated by phosphorylation of two sites, serine-112 (Ser-112) and serine-136 (Ser-136). Phosphorylation at either site results in loss of the ability of BAD to heterodimerize with the survival proteins BCL-XL or BCL-2. Phosphorylated BAD binds to 14-3-3 and is sequestered in the cytoplasm. (PMID: 10597268)
