  WB analysis using 84906-1-RR (same clone as 84906-1-PBS)

  Non-treated A431 cells and EGF (HZ-1326) treated A431 cells were subjected to SDS PAGE followed by western blot with 84906-1-RR (Phospho-EGFR (Tyr1197) antibody) at dilution of 1:2000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Alpha Tubulin (66031-1-Ig) antibody as a loading control. This data was developed using the same antibody clone with 84906-1-PBS in a different storage buffer formulation.

  FC experiment of A431 using 84906-1-RR (same clone as 84906-1-PBS)

  1x10^6 A431 cells untreated (dashed lines) or treated with EGF which intracellularly stained with 0.25 ug Phospho-EGFR (Tyr1197) Recombinant Antibody (84906-1-RR, Clone:241826G10) and PE-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with 90% MeOH. This data was developed using the same antibody clone with 84906-1-PBS in a different storage buffer formulation.

Background Information

Epidermal Growth Factor Receptor (EGFR) is one of the receptor tyrosine kinases (RTKs), members of the ErbB/HER family which consists of ErbB1 (EGFR or HER1), ErbB2 (HER2 or Neu), ErbB3 (HER3), and ErbB4 (HER4). The EGFR autophosphorylation sites Tyr-992 and Tyr-1173 (also known as Tyr 1197) have been shown to play a critical role in the activation of the MAPK cascade following EGF stimulation. (PMID: 15708852) Some scientists call EGFR Tyr1197 as EGFR Tyr1173, the site where the signaling peptide is removed.
