Background Information
IRAK4 is the central kinase in the Toll-like receptor pathway of the innate immune system. Toll-like receptors constitute the first line of defense against pathogenic micro-organisms such as bacteria, viruses, and yeast. Unphosphorylated full-length IRAK4 can autophosphorylate in vitro in a cell-free system. In the cellular context, however, autophosphorylation is inducible by IL-1β. Four phosphorylation sites have been identified in the activation loop in some articles, which are the results of autophosphorylation: Thr-342, Thr-345, Ser-346 and Thr-352. (PMID: 24567333)
