  WB analysis of HEK-293T using 81398-1-RR (same clone as 81398-1-PBS)

  Non-treated HEK-293T cells, phosphatase inhibitor and λ phosphatase treated HEK-293T cells were subjected to SDS PAGE followed by western blot with 81398-1-RR (Phospho-MNK1 (Thr250/255) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with GAPDH antibody as loading control. This data was developed using the same antibody clone with 81398-1-PBS in a different storage buffer formulation.

  WB result of Phospho-MNK1 (Thr250/255) antibody (81398-1-RR; 1:2000; incubated at room temperature for 1.5 hours) with sh-Control and sh-Phospho-MNK1 (Thr250/255) transfected HEK-293 cells. This data was developed using the same antibody clone with 81398-1-PBS in a different storage buffer formulation.

Background Information

MNK1/MKNK1 (MAPK Interacting Serine/Threonine Kinase 1) is a Ser/Thr protein kinase that interacts with, and is activated by ERK1 and p38 mitogen-activated protein kinases, and thus may play a role in the response to environmental stress and cytokines. MNK1 regulates diverse biologic processes including translation, cell proliferation, and differentiation through phosphorylation of different substrates(PMID: 29045529). Through phosphorylation MNK1 regulates the function of eukaryotic translation initiation factor 4E (eIF4E), a key player in translational control, whose expression is mostly upregulated in cancer patients.
