  WB analysis using 86641-1-RR

  Non-treated ACHN cells and IL-4 (HZ-1004) treated ACHN cells were subjected to SDS PAGE followed by western blot with 86641-1-RR (Phospho-STAT6 (Tyr641) antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours. The membrane was stripped and re-blotted with Vinculin (66305-1-Ig) antibody as a loading control.

  IF Staining of ACHN using 86641-1-RR

  Immunofluorescent analysis of (4% PFA) fixed IL-4 treated ACHN cells using Phospho-STAT6 (Tyr641) antibody (86641-1-RR, Clone: 250940C12 ) at dilution of 1:500 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-Phalloidin (red).

  FC experiment of ACHN using 86641-1-RR

  1X10^6 ACHN cells untreated (dashed lines) or treated with IL-4 treated which intracellularly stained with 0.13 ug Phospho-STAT6 (Tyr641) Recombinant monoclonal antibody (86641-1-RR, Clone:250940C12) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.13 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed and permeabilized with Flow Cytometry Phosphorylated Protein Fixation/Permeabilization Kit (PF00026).

Background Information

STAT6, also named as Signal transducer and activator of transcription 6, is a 847 amino acid protein, which belongs to the transcription factor STAT family. p-STAT6(Tyr641) is a biomarker of Th2-high inflammation, targeted by IL-4Rα blockers (dupilumab) and JAK1/3 inhibitors. High p-STAT6 in TAMs correlates with M2 polarization and radioresistance, blocking IL-4 or JAK1/3 decreases Tyr641 phosphorylation and restores anti-tumor immunity. (PMID: 37474750)
