  WB analysis using 86740-1-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 86740-1-RR (RNF43 antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  IF Staining of HEK-293 using 86740-1-RR

  Immunofluorescent analysis of (-20°C Ethanol) fixed HEK-293 cells using RNF43 antibody (86740-1-RR, Clone: 251708C1 ) at dilution of 1:400 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-Phalloidin (red).

Tested this RNF43 antibody with overexpressed RNF43 tagged with BromoTag-GFP-HA and couldn't see any bands at the correct molecular weight. I have also included the HA I blotted to show RNF43. It seems the antibody binds to something non-specific and it isn't at the right molecular weight to be endogenous RNF43. With RNF43 being overexpressed, bands should be stronger than endogenous level, but couldn't be detected. Also, tags are on the C-terminus which shouldn't affect binding of the antibody with the antibody binding domain being at 24-197 aa
