  WB analysis using 31485-1-AP (same clone as 31485-1-PBS)

  U-937 cells were subjected to SDS PAGE followed by western blot with 31485-1-AP (SIRP Alpha antibody) at dilution of 1:1500 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 31485-1-PBS in a different storage buffer formulation.

  IF Staining of THP-1 using 31485-1-AP (same clone as 31485-1-PBS)

  Immunofluorescent analysis of (4% PFA) fixed THP-1 cells using SIRP Alpha/CD172a antibody (31485-1-AP) at dilution of 1:200 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2). This data was developed using the same antibody clone with 31485-1-PBS in a different storage buffer formulation.

  FC experiment of THP-1 using 31485-1-AP (same clone as 31485-1-PBS)

  1x10^6 THP-1 cells were intracellularly stained with 0.25 ug SIRP Alpha/CD172a Polyclonal antibody (31485-1-AP) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Rabbit IgG Isotype Control RecAb (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer. This data was developed using the same antibody clone with 31485-1-PBS in a different storage buffer formulation.

Background Information

SIRP Alpha, also known as CD172a, SHPS-1, and BIT, belongs to the SIRP family. SIRP Alpha is a transmembrane glycoprotein expressed explicitly on myeloid cells and provides a "do not eat me" signal after engaging with integrin-associated protein CD47 on tumor cells (PMID: 36419386). SIRP Alpha shows heterogeneity in molecular weight (~65-120 kDa) in various tissues due to differential glycosylation (PMID: 18051954).
