Background Information

TMEM120A (also known as TACAN) is a nuclear envelope transmembrane protein essential for adipocyte differentiation and lipid metabolism. Originally identified through proteomic screening, it regulates spatial genome organization by repositioning pro-adipogenic genes from the nuclear periphery to the interior during adipogenesis. Adipocyte-specific knockout mice exhibit lipodystrophy with insulin resistance. TMEM120A was initially proposed as a mechanosensitive ion channel involved in mechanical pain sensing; however, subsequent structural studies revealed it contains a coenzyme A-binding site and shares homology with fatty acid elongases, challenging its role as an ion channel (PMID: 26024229; 34409941; 34374645).
