  WB analysis using 84711-5-RR

  Various lysates were subjected to SDS PAGE followed by western blot with 84711-5-RR (TOLLIP antibody) at dilution of 1:10000 incubated at room temperature for 1.5 hours.

  IHC staining of mouse brain using 84711-5-RR

  Immunohistochemical analysis of paraffin-embedded mouse brain tissue slide using 84711-5-RR (TOLLIP antibody) at dilution of 1:400 (under 40x lens). Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0).

  IF Staining of HepG2 using 84711-5-RR

  Immunofluorescent analysis of (-20°C Ethanol) fixed HepG2 cells using TOLLIP antibody (84711-5-RR, Clone: 242098E3 ) at dilution of 1:400 and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2), CL594-Phalloidin (red).

  FC experiment of U2OS using 84711-5-RR

  1x10^6 U2OS cells were intracellularly stained with 0.25 ug TOLLIP Recombinant antibody (84711-5-RR, Clone:242098E3) and PE-Conjugated Goat Anti-Rabbit IgG(H+L)(red), or 0.25 ug Rabbit IgG Isotype Control Recombinant Antibody (98136-1-RR, Clone: 240953C9) (blue). Cells were fixed with 4% PFA and permeabilized with Flow Cytometry Perm Buffer.

  Affinity and Kinetic Characterization of 84711-5-RR

  Biolayer interferometry (BLl) kinetic assays of 84711-5-RR against Human TOLLIP were performed. The affinity constant is 0.309 nM.

Background Information

TOLLIP (Toll interacting protein), is an inhibitory adaptor protein. Tollip is a negative regulator of IL-1R and Toll-like receptors signalling pathway, involved in inflammatory immune response and inhibiting IRAK1 phosphorylation and kinase activity (PMID:11441107, PMID:10854325, PMID:11751856). The TOLLIP-dependent selective autophagy pathway plays a role in clearance of cytotoxic polyQ proteins aggregates (PubMed:25042851).
