Background
IL-10 is a non-covalent homodimeric glycoprotein with an apparent molecular mass of 17 kDa. IL-10 is produced by a variety of mammalian cell types including T cells, B cells, macrophages, and keratinocytes monocytes. It is an anti-inflammatory cytokine with multiple roles in inflammation and immunoregulation (PMID: 19161426; 9162098).

FH Julie (Verified Customer) (10-23-2025) | Used in cell culture to polarise differentiated THP-1 cells to M2c phenotype. IL-10 was reconstituted and added to RPMI + Glutamax with 10% FBS and 1% PenStrep at a concentration of 20ng/mL with 1 x 10^6 cells/mL. Cells were then incubated for 48hrs with no media change, and CD163 (unconjugated + Coralite Plus 488 secondary, Proteintech) was used in flow cytometry to confirm polarisation.
