Background Information
EIF2AK4 (eukaryotic translation initiation factor 2 α kinase 4), formerly known as GCN2, a member of serine/threonine kinases, is comprised of an N-terminal RWD domain (20-137) involved in protein-protein interactions, pseudokinase (276-539) and kinase domains (585-1016) and C-terminal domain (1533-1649) where binding to ribosomes and recognition of uncharged tRNAs occur. It downregulates protein synthesis in response to a variety of cellular stress such as hypoxia, viral infection, and amino acid deprivation (PMID: 38776952, 26300654). Mutant EIF2AK4 could promote proliferation of small pulmonary arteries by crosstalk with mammalian targets of the rapamycin signaling pathway (PMID: 24135949).
