Background Information

Golgi-associated, gamma-adaptin ear-containing, ARF-binding protein 3 (GGA3) is a monomeric clathrin adaptor that localizes predominantly to the trans-Golgi network (TGN) and endosomal membranes. Beyond classical cargo sorting, GGA3 participates in receptor recycling, retrograde transport and signal termination. Notably, caspase-3 cleaves GGA3 during apoptosis or cerebral ischaemia; loss of this adaptor stabilises β-secretase (BACE1), thereby increasing amyloid-β production and linking GGA3 to Alzheimer disease pathogenesis. Recent work further implicates GGA3 in oncogenic processes: in non-small-cell lung cancer it supports TrkA trafficking to the plasma membrane, prolonging Akt/ERK signalling and promoting cell survival.(PMID: 39267722, PMID: 37586201)
