The ChromoTek His Fab-Trap® Magnetic Particles M-270 Kit consists of an anti-His-tag Fab-fragment, which is covalently bound to magnetic particles. It also contains lysis, wash, and elution buffers that can be used for the immunoprecipitation of His-tagged proteins from cell extracts of various organisms. Specificity/TargetBinds specifically to the His-tag (also known as hexahistidine or His6-tag) fused to a protein of interest at N-, C- or internal position. Please note that the affinity is highest for a C-terminal fusion (dissociation constant KD of 10 nM for a C-terminal His-tag and 220 nM for an N-terminal His-tag).

We have a reasonably large complex that I have been trying to purify using more classical protein purification approaches, however it is sensitive to buffer changes and was falling apart or degrading. I was recommended this kit by our account manager Jess along with the his-peptide (hp-1) to use as a milder elution, compared to the classical acid-based elutions. The downstream application of this is structural analysis by cryo-EM and we therefore wanted to maintain the arrangements. This kit worked really well extracting our his-tagged complex from cells at a high enough concentration and purity for cryo-ET, and with the complexes still intact.
