  WB analysis using 84899-5-RR (same clone as 84899-5-PBS)

  Various lysates were subjected to SDS PAGE followed by western blot with 84899-5-RR (PSPH antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 84899-5-PBS in a different storage buffer formulation.

  FC experiment of HepG2 using 84899-5-RR (same clone as 84899-5-PBS)

  1x10^6 HepG2 cells were intracellularly stained with 0.25 ug PSPH Recombinant monoclonal antibody (84899-5-RR, Clone:242518C4) and CoraLite®488-Conjugated Goat Anti-Rabbit IgG(H+L) (SA00013-2)(red), or 0.25 ug Isotype Control (blue). Cells were fixed and permeabilized with Intracellular Flow Cytometry Fixation & Permeabilization Buffer Kit (PF00019). This data was developed using the same antibody clone with 84899-5-PBS in a different storage buffer formulation.

  Affinity and Kinetic Characterization of 84899-5-RR

  Biolayer interferometry (BLl) kinetic assays of 84899-5-RR against Human PSPH were performed. The affinity constant is below 1 pM.

Background Information

PSPH (phosphoserine phosphatase) is an enzyme, which is involved in the process of L-serine biosynthesis. PSPH mainly plays role in multiple aspects of cell behaviours such as proliferation and differentiation by producing precursors for the biosynthesis of diverse compounds including neurotransmitters, glycolipids and thymidine (PMID: 11237721, PMID: 19963421). Additionally, augmented PSPH level is correlated with the prognosis in multiple cancers including cutaneous squamous cell carcinoma (PMID: 21726982), breast cancer (PMID: 28931725), non-small cell lung cancer (PMID: 30662358), colorectal cancer (PMID: 24146633) and hepatocellular carcinoma (PMID: 25793315).
