Background Information
RASAL2 (RAS protein activator like 2) is a protein-coding gene that encodes a member of the RAS GTPase-activating protein (RAS-GAP) family. The encoded protein contains a characteristic GAP-related domain (GRD) that enables it to catalyze the hydrolysis of RAS-GTP to RAS-GDP, thereby functioning as a negative regulator of RAS signaling. Structurally, RASAL2 also possesses additional domains including a Pleckstrin homology (PH) domain and a C2 domain, which are involved in membrane localization and calcium/lipid binding. RASAL2 has been extensively studied for its context-dependent roles in cancer, predominantly functioning as a tumor suppressor in malignancies such as breast, bladder, renal cell, ovarian, and lung cancers, where its downregulation or loss promotes tumor growth, stemness, epithelial-mesenchymal transition (EMT), and metastasis.
