  WB analysis of K-562 using 67948-1-Ig (same clone as 67948-1-PBS)

  K-562 cells were subjected to SDS PAGE followed by western blot with 67948-1-Ig (TNIK antibody) at dilution of 1:5000 incubated at room temperature for 1.5 hours. This data was developed using the same antibody clone with 67948-1-PBS in a different storage buffer formulation.

Heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0). This data was developed using the same antibody clone with 67948-1-PBS in a different storage buffer formulation.

  Immunofluorescent analysis of (4% PFA) fixed A431 cells using TNIK antibody (67948-1-Ig, Clone: 1E9H3 ) at dilution of 1:800 and CoraLite®488-Conjugated AffiniPure Goat Anti-Mouse IgG(H+L), CL594-Phalloidin (red). This data was developed using the same antibody clone with 67948-1-PBS in a different storage buffer formulation.

  FC experiment of A431 using 67948-1-Ig (same clone as 67948-1-PBS)

  1X10^6 A431 cells were intracellularly stained with 0.4 ug Anti-Human TNIK (67948-1-Ig, Clone:1E9H3) and CoraLite®488-Conjugated AffiniPure Goat Anti-Mouse IgG(H+L) at dilution 1:1000 (red), or 0.4 ug Mouse IgG2a Isotype Control (C1.18.4) (65208-1-Ig, Clone: C1.18.4) (blue). Cells were fixed and permeabilized with Transcription Factor Staining Buffer Kit (PF00011). This data was developed using the same antibody clone with 67948-1-PBS in a different storage buffer formulation.

Background Information

Traf2- and Nck-interacting kinase (TNIK) is one of the germinal center kinase family members involved in cytoskeleton organization and neuronal dendrite extension (PMID: 24566388). TNIK is an essential regulatory component of the T-cell factor-4 and β-catenin transcriptional complex and is required for the tumor-initiating function of colorectal cancer stem cells (PMID: 10521462).Western blot analysis detected TNIK at an apparent molecular mass of 150~180 kDa as cited in literature (PMID: 24566388).
